Differences in distribution pattern of marker enzymes among subcellular fractions from Morris hepatoma 16.

چکیده
The distribution of marker enzymes in subcellular frac tions isolated by differential centrifugation was determined in Morris hepatoma 16, host liver, and control rat liver. Marker enzymes of cellular organdÃ-esthat were assayed in cluded cytochrome oxidase, monoamine oxidase, ornithine aminotransferase, D-amino acid oxidase, acid phosphatase, glucose 6-phosphatase, ethylmorphine demethylase, and lactic dehydrogenase. Comparison of host liver and hepa toma revealed significant differences in the distribution pattern of many of these enzymes, especially those asso ciated with mitochondria. The hepatoma showed increased amounts of mitochondria! enzyme activity in a subcellular fraction requiring higher g forces than normal for sedimen tation. This could mean that hepatoma 16contains a greater proportion of mitochondria that are smaller in size and/or less dense than those of host or normal liver. Electron micro graphs of hepatoma and host liver tissue and subcellular fractions also suggested that hepatoma 16 contains mito chondria that are smaller in size. It is concluded, therefore, that a greater proportion of mitochondria of hepatoma 16 are smaller in size than those of host liver.

متن کاملMICROSOME-MEDIATED BENZO[A]PYRENE-DNA BINDING AND INHIBITION BY CYTOSOLIC FRACTIONS FROM LIVER AND SKIN OF ADULT AND WEANLING RATS
Biotransformation of benzo[a]pyrene (BaP) in the presence of microsomal fractions derived from liver and epiderm of adult and weanling rats was examined. The aim of this study was to evaluate the effect of age on the capacity of two organs in transformation of BaP. Subcellular fractions were prepared from skin and liver by ultracentrifugation and were used as the source of BaP metabolizing enzy...
متن کاملSome biochemical characteristics of rat liver and Morris hepatoma nuclei and nuclear membranes.
Nuclei prepared from host liver and from Morris hepatomas 7777 and 7800 have been compared with respect to some of their biochemical characteristics. Several criteria were used to ensure that liver and hepatoma nuclei were of equal purity. These criteria include equal specific activity ratios (homogenate nuclei) for several marker enzymes. Phospholipids, proteins, and sialic acid content were c...
متن کاملAltered subcellular and submitochondrial localization
The subcellular and submitochondrial localization of CTP:phosphatidate cytidylyltransferase is altered in the Morris 7777 hepatoma. Mitochondria in this poorly differentiated tumor are the principal sites of CDP-diacylglycerol synthesis, in contrast to normal rat liver where the endoplasmic reticulum is most active. This enzyme activity was increased 1 7-fold in the outer mitochondrial membrane...
متن کاملCharacterization of subcellular components in synchronized hepatoma cells as a function of the cell cycle.
The specific activity and subcellular distribution of marker enzymes for the main subcellular components were analysed in homogenates of synchronized hepatoma cells (Morris 7288c), obtained by selective detachment at mitosis combined with a metaphase block with Colcemid. Markers for lysosomes, mitochondrial outer membrane, plasma membrane and cytosol are synthesized throughout the cycle at the ...
